Golimumab : a new anti - TNF agent on the horizon for infl ammatory arthritis
نویسنده
چکیده
Rheumatoid arthritis (RA), psoriatic arthritis, and ankylosing spondylitis are highly prevalent infl ammatory arthritides. Various conventional therapies are currently in use, and biologic therapies have also been introduced into the treatment armamentarium. Despite the remarkable effi cacy of the anti-tumor necrosis factor (TNF) agents, there still remains an unmet need for newer anti-TNF agents. A substantial number of patients do not respond to an initial anti-TNF agent, but may respond to a second one. The modes of administration of currently marketed anti-TNF agents are not particularly convenient and risk-free for all patients. An anti-TNF agent with a better safety profi le is also highly desirable. Golimumab (CNTO-148) is a high affi nity, human monoclonal antibody to TNF-α. The various ongoing trials for golimumab have shown promising results in terms of effi cacy and safety in methotrexate-naive and -resistant patients with RA, as well as in patients with RA who were previously treated with other anti-TNF agents. In addition, the effi cacy of golimumab in psoriatic arthritis and ankylosing spondylitis has also been demonstrated.

RHEUMATOID ARTHRITIS IS AN AUTOIMMUNE disease that affects 1.3 million adults in the United States.1 Of these, about 75 percent are women. Disease onset generally occurs between 30 and 50 years of age. Disease hallmarks are synovial infl ammation, progressive bone erosion, joint mal-alignment and destruction, and subsequent weakness of surrounding tissues and muscles. Systemic manifestations al...
متن کاملAnti-inflammatory Effects of Yopm in Rheumatoid Arthritis Fibroblast-like Synoviocytes
Introduction Fibroblast-like synoviocytes (FLS) are key players in the pathogenesis of rheumatoid arthritis (RA). During the onset of the disease, they are transformed into an aggressive phenotype that sustains the joint-destructive infl ammatory environment of RA. This transformation is most likely due to the combination of cytokines, growth factors and cellular interaction, which leads to the...

W ith increasing curiosity into more holistic approaches to disease prevention and treatment, some researchers are rediscovering how an ancient spice may help treat and prevent many types of cancer. Curcumin, or diferuloylmethane, is a yellow-orange powder that is the principal ingredient in turmeric (Curcuma longa), the Indian spice popularly known as curry. and other parts of Asia and tropica...

not eff ective for many persons with rheumatoid arthritis (RA) and other infl ammatory rheumatic diseases. At this time RA patients had a signifi cantly decreased life expectancy due to a relatively incontrolled infl ammatory activity of RA combined with an increased incidence of comorbidities and severe treatment-related side eff ects. After 5 years of disease duration, a considerable percenta...
متن کاملAnti-TNF-α therapy modulates resistin in patients with rheumatoid arthritis
Objective Chronic systemic infl ammation plays a pivotal role in the development of atherosclerosis in rheumatoid arthritis (RA). In the present study, we investigated whether anti-TNF-α antagonist-monoclonal antibody-infl iximab administration alters circulating levels of resistin, a proinfl ammatory adipokine. We further assessed associations of circulating resistin concentrations with CRP an...
